A comparison between enzyme immunoassay and HPLC for ochratoxin A detection in green, roasted and instant coffee BABT

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for ochratoxin A (OTA) detection in green, roasted and instant coffees was developed using anti-OTA monoclonal antibody. Immunological reagents prepared were OTA-BSA (4.76 mg/mL), anti-OTA.7 MAb (2x10³-fold dilution) and HRP-anti IgG (10³-fold dilution). The detection limit was 3.73 ng OTA/g and correlation coefficients (r) between this immunoassay and high performance liquid chromatography were 0.98 for green coffee, 0.98 for roasted and 0.86 for instant. OTA levels detected by ic-ELISA were higher than by HPLC, with ELISA/HPLC ratio of 0.66 - 1.46 (green coffee), 0.96 - 1.11 (roasted) and 0.93 - 1.82 (instant). This work investigated the effects of co-occurring aflatoxin B1 (AFB1) and microcystin (MC) in aquaculture, using immunohistochemistry and genotoxicity methods. Tilapia (Oreochromis niloticus) were exposed to AFB1 by intraperitoneal and MC (cell extract of Microcystis aeruginosa) by intraperitoneal and immersion routes. The interaction of MC-AFB1 was evaluated co-exposing the intraperitoneal doses. Blood samples were collected after 8, 24, and 48h to analyze the micronucleus frequency and comet score. The interaction of MC-AFB1 showed a synergic mutagenic response by higher micronucleus frequency of co-exposed group. A slight genotoxic synergism was also observed in the comet score. Fruits from temperate and tropical climates which have high levels of antioxidant compounds are the source of numerous studies concerning the correlation with benefits to human health. The objectives of this study were to quantify the anthocyanins and phenolic compounds and also to measure the antioxidant activity (ferric reducing antioxidant power - FRAP) of blackberries from two varieties grown in southern Brazil ('Brazos' and 'Tupy') at three stages of ripening; unripe, semi-ripe, ripe and their products (pulp and fermented products). Comparison of thin-layer chromatography, spectrofluorimetry and bright greenish-yellow fluorescence test for aflatoxin detection in corn BABT

In this study the bright greenish-yellow fluorescence test, widely used by the corn milling industry, was compared to the thin-layer chromatography (TLC) and spectrofluorimetry methods for aflatoxin detection in 40 corn samples naturally contaminated by the Aspergillus section Flavi. According to the corn processing industry criteria, all the samples were adequate for human and animal consumption by the bright greenish-yellow fluorescence test, but TLC and spectrofluorimetry analysis detected aflatoxins above the maximum tolerated limit (20 µg/kg) in 7 and 8 samples, respectively. Culture medium for amylase production by toxigenic fungi BABT

Mycelial growth and amylase production by a mycotoxigenic strain of Fusarium moniliforme and Aspergillus flavus were evaluated in a culture medium containing starch, glycerol, wheat bran or corn. With emphasis on corn, different fractions composed by germ, degermed seed, starch, milky stage corn and the respective starch or supernatant fraction were analyzed for F. moniliforme growth . The medium composed of milky stage corn supernatant promoted the best mycelial growth (p<0.05), and it was used to prepare amylase production medium in the next step. Abstract: The objective of this work was to standardize and validate an indirect competitive enzyme-linked immunosorbent assay (ic-Elisa), as a low-cost tool, to monitor the presence of aflatoxin in common and blanched peanuts (Arachis hypogaea) in the production chain. The presence of aflatoxin B1, moisture content, and water activity were analyzed in 60 samples of the peanut cultivar Runner IAC 886, from the 2014/2015 and 2015/2016 harvests of the region of Alta Paulista, in the state of São Paulo, Brazil. Fumonisins in corn: correlation with Fusarium sp. count, damaged kernels, protein and lipid content BABT

Natural fungal and fumonisin contamination were evaluated in 109 freshly harvested corn samples from Paraná State and correlated to damaged kernels (%). In addition, healthy and damaged kernels of 24 corn samples were selected in order to compare the mycoflora profile and fumonisin levels. The correlation among protein/lipid content and fumonisin levels was also analyzed in the 15 most frequently cultivated corn hybrids. Total fungal colony count in 109 freshly harvested corn samples ranged from 1.9x10(4) to 3.5x10(6) CFU/g, Fusarium sp. count from 1.0x10³ to 2.2x10(6) CFU/g, and fumonisin levels from 0.13 to 20.38 µg/g. Fusarium verticillioides strains isolated from corn feed: characterization by fumonisin production and RAPD fingerprinting BABT

In this study a total of 16 Fusarium verticillioides strains isolated from corn feed samples were characterized by fumonisin (FB) production and random amplified polymorphic DNA (RAPD). All the strains produced FB1 and FB2 with levels ranging from 2.41 to 3996.36 µg/g, and from 1.18 to 1209.91 µg/g, respectively. From the 16 F. verticillioides strains, four were identified as low (3.59 to 1289.84 µg/g), eight as intermediate (>1289.84 to 3772.44 µg/g) and four strains as high (>3772.44 µg/g) fumonisin producers. Low cost production and purification of polyclonal antibodies to staphylococcal enterotoxin A Rev. Microbiol.

An immunization scheme for production of antiserum to staphylococcal enterotoxin A (SEA) is proposed. The reference method of Robbins and Bergdoll was modified to reduce the number of doses and the amount of toxin used per animal. The best immunization scheme used injections in days 0, 8, 24, 59, 62 and 67. The amount of toxin at each injection was 5, 6, 20, 50, 100 and 200<FONT FACE="Symbol">m</font>g, respectively. The total amount of toxin was 381<FONT FACE="Symbol">m</font>g, which corresponded to a reduction of 107<FONT FACE="Symbol">m</font>g in the amount of toxin for each animal when compared to the reference method. Penicillium expansum versus antagonist yeasts and patulin degradation in vitro BABT

Taking into account the preliminary antagonistic/biodegradation property showed by Pichia membranifaciens and Sporobolomyces roseus, which decreased the initial patulin concentration of 588.4 to 290.0 µg/mL, ability of P. ohmeri 158 in biocontrol against Penicillium expansum and patulin decrease in vitro was performed. The culture supernatant of P. ohmeri 158 was effective against 66.17% micelial growth, indicating antibiosis related with the killer phenomenon. The initial patulin concentration of 223 µg in the presence of P. ohmeri 158 cells was decreased over 83% of the original concentration, when incubated at 25ºC/2 days and > 99% after 5 days incubation time, with undetectable patulin level after 15 days.
